Asset Overview

This model demonstrates some of the key aspects of the E47:SCL Helix-Loop-Helix Heterodimer. This heterodimer forms a multiprotein complex with the LMO2:LDB1 bridging proteins which also affects the hydrogen bonding occurring in E47:SCL. This complex plays a role in the human body to regulate haematopoiesis and induces T-cell Leukaemia. The structure above represents the E47:SCL complex bound to the major groove of an E-box DNA motif 5'-C1-A2-G3-A4-T5-G6-3' (forward). The reverse strand is annotated with a * The heterodimer contains some key highly conserved residues such as V595 (shown in red) which impact the formation of salt bridges and other interactions which are indicative of the roles and structures of similarly classed HLH protein complexes. The structure above uses a colour code where SCL=cream helices, with yellow indicating the loop; E47=light blue helices with dark blue indicating the loop; the E-box DNA=dark grey.
